Extracellular RNA (exRNA): Structure, Functions, and Emerging Applications

A study published in the journal ‘Clean Water’ reported the presence of extracellular RNA (exRNA) from bacteria in disinfected drinking water.

  • Analysis of exRNA helps scientists understand bacterial activity just before death, revealing survival strategies.
    • These insights can be used to develop more effective disinfectants

About Extracellular RNA (exRNA)

  • exRNA is RNA that exists outside cells, in body fluids such as blood, saliva, urine, and cerebrospinal fluid.
  • Unlike intracellular RNA, exRNA functions as a communication molecule between cells.

Key Features

  • Mode of Release: Extracellular RNA (exRNA) is released by cells either through active secretion or as a result of cell damage and death.
  • Structural Packaging: exRNA is typically enclosed within extracellular vesicles (EVs), such as exosomes, or remains bound to proteins and lipids.
  • Protection Mechanism: These vesicles and molecular associations protect exRNA from degradation by ribonucleases (RNases) present in the extracellular environment.
  • Long-Distance Transport: exRNA has the ability to travel long distances within the body, enabling intercellular communication across different tissues.

Mechanism of Action of Extracellular RNA (exRNA)

  • Release into Extracellular Space: Cells release extracellular RNA (exRNA) into the surrounding extracellular environment.
  • Mode of Transport: exRNA is transported through extracellular vesicles or remains associated with protein complexes.
  • Cellular Uptake: Recipient or target cells take up the exRNA from the extracellular space.
  • Functional Impact: exRNA modifies gene expression and influences cellular behaviour in the target cells.

Applications

  • Non-Invasive Biomarker: Extracellular RNA (exRNA) can be utilized as a non-invasive biomarker through techniques such as liquid biopsy.
  • Early Disease Detection: It aids in the early detection of diseases, including cancer.
  • RNA-Based Treatment: exRNA holds potential for use in RNA-based drugs and targeted therapeutic interventions.
  • Microbial Activity Indicator: exRNA present in water can help reveal microbial activity and their survival strategies.
  • Communication Role: exRNA acts as a cell-to-cell signaling molecule.
  • Gene Regulation: It modifies gene expression in recipient cells.

Limitations

  • Technical Complexity: Isolation and analysis of exRNA are difficult due to low concentration and contamination.
    • For Example: Separating exRNA from blood plasma without contamination from cellular RNA remains a major laboratory challenge.
  • Lack of Standardization: There are no universally accepted protocols for extraction, storage, and analysis.
    • For Example: Different studies on cancer biomarkers using exRNA often give inconsistent results due to varying methodologies.
  • Stability Variability: Although protected, exRNA stability varies depending on conditions and carriers.
    • For Example: exRNA in improperly stored blood samples may degrade, affecting diagnostic accuracy.
  • Complex Data Interpretation: It is difficult to determine the exact source and function of exRNA.
    • For Example: A specific microRNA detected in blood may originate from multiple tissues, making disease identification ambiguous.
  • High Cost and Infrastructure Needs: Advanced tools and expertise are required for exRNA research.
    • For Example: Techniques like next-generation sequencing (NGS) for exRNA profiling are expensive and not widely available in developing countries.
